川楝子致小鼠肝毒性时效、量效关系研究

摘    要:目的研究川楝子致小鼠肝毒性的时效、量效关系。方法小鼠灌服不同剂量川楝子,测定其LD50。另取小鼠按不同时间点或不同剂量分组,检查血清ALT,AST,计算肝、脾、胸腺脏器指数,并观察肝组织形态变化。结果小鼠灌服川楝子的LD50为200 g.kg-1。ALT,AST在给药后1 h达到高峰,24 h可恢复近正常值。与正常组比较,川楝子81,166 g/kg剂量组对肝组织产生明显损伤,且随着剂量增大,ALT,AST升高显著(P<0.05或P<0.01)。结论小鼠单次灌服一定剂量川楝子可造成急性肝损伤甚至死亡,并显示有毒性时效、量效关系。
