奥沙利铂诱导survivin表达改变与宫颈癌细胞化疗敏感性的关系

目的探讨survivin基因在奥沙利铂（OXA）诱导的宫颈瘸细胞凋亡中的作用。方法用宫颈癌Hela细胞株进行培养传代，MTT试验检测OXA不同药物浓度对宫颈癌细胞生长的抑制作用。分别在加药前、加药后24、48、72和96h采用逆转录聚合酶链反应（RT-PCR）检测survivin mRNA的表达。结果OXA对宫颈癌细胞生长均有明显的抑制作用。并有浓度和时间的依赖性。随浓度增加。宫颈癌细胞凋亡率表现先上升，后下降；随时间延长，在高浓度OXA组凋亡率明显增加，在低浓度OXA组凋亡率增加不明显。0．1mg／L和1mg／L OXA处理宫颈癌细胞，survivin mRNA表达随作用时间的延长而增加。结论小剂量OXA处理宫颈瘸细胞，stirvivin mRNA表达随作用时间的延长而增加，这可能是宫颈癌细胞对化疗药物产生耐药性的重要因素之一。

The Role of Survivin in the Apoptosis of Cervical Cancer Cell Line Induced by Oxaliplatin

Objective To study the role of survivin in the apoptosis of cervical cancer cell line induced by oxaliplatin(OXA).Methods The inhibition effects of different doses of OXA on Hela cell line were assayed with MTT test.Cultured Hela cells were divided into 2 groups: OXA and control.OXA group was divided into 10,4,2,1,0.1,0.01 mg/L groups.The expression of survivin was determined by RT-PCR on 24,48,72and 96 h after Hela cells exposed to oxaliplatin.Results OXA obviously inhibited the cervical cancer cell growth a manner of dose dependent and time-dependent.Apoptosis induced by chemotherapeutical drug increased at first and then decreased as the concentrations of OXA got higher while as the time went by,the apoptosis in the high OXA concentration group increased significantly but in the low OXA concentration group increased slowly.As time went on,survivin mRNA level was increased after Hela cells exposed to 0.1 mg/L and 1 mg/L of OXA.Conclusions A high expression of survivin was induced in Hela cells by low concentration of oxaliplatin in a time-dependent manner,which may be caused by chemoresistence of cervical ealinonla cells induced by chemotherapy agent.
