摘    要:观察尾叶香茶菜中提取物二萜类化合物F对人肝癌细胞株SMMC-7721的作用并探讨其机制。分别应用MTT法、AO/EB荧光染色法分析细胞存活率和凋亡情况,利用DCFH-DA荧光分光光度法检测细胞内过氧化物(ROS)的水平。二萜类化合物F抑制SMMC-7721细胞增殖,且呈时间剂量依赖性,二萜类化合物F作用的SMMC-7721细胞内ROS水平明显升高。二萜类化合物F可以启动细胞内氧化应激诱导SMMC-7721细胞坏死或凋亡而抑制肝癌细胞株增殖。

Abstract:To determine the apoptosis of human hepatoma carcinoma cell line SMMC -7721 induced by diterpenoid F derived from radbosia excise, and to study its mechanism. The survival and apoptotic rate of SMMC -7721 cells were detected by MTT and AO/EB fluorescent staining respectively. And the intracellular ROS level was analyzed by fluorospectro-photometry. Diterpenoid F could inhibit the proliferation of SMMC -7721 in a dose - and time - dependent manner. The intracellular ROS increases in SMMC -7721 through oxidative stress pathway.
