大豆黄酮对肝癌SMMC7721细胞及其CD~(133)肿瘤干细胞增殖的影响

摘    要:目的:探讨大豆黄酮对肝癌SMMC7721细胞及其CD133肿瘤干细胞增殖的影响。方法:取对数生长期的肝癌SMMC7721细胞,分别加入不同浓度的大豆黄酮,在24、48和72 h的时间点上,用MTT法测定大豆黄酮对肝癌SMMC7721细胞的抑制生长率。另外,将培养的细胞与CD133蛋白荧光素一抗标记混合,37℃孵育反应20 min,在525 nm波长下进行流式细胞仪检测。结果:大豆黄酮对于肝癌SMMC7721细胞在24、48和72 h内均有明显的抑制作用,并且抑制率随浓度的增加而增加。经大豆黄酮处理24 h后,CD133肿瘤干细胞的数量也显著减少。结论:大豆黄酮不仅具有抑制肿瘤细胞增殖的作用,而且也具有抑制CD133肿瘤干细胞增殖的作用。

Abstract:Objective: In order to explore the influence of soybean flavones to the proliferation of liver cancer SMMC7721 cell lines and CD133 tumor stem cells.Methods: Liver cancer SMMC7721 cell lines are cultured to logarithmic growth time,and the soybean flavones of deferent concentrations were added into the media.After the culture of 24 h,48 h and 72 h,the inhibiting rates of soybean flavones to liver cancer SMMC7721 cells were measured by MTT method.In addition,the cultured cells were mixed up with CD133 antibody marked with fluorescein.After culturing at 37 ℃ for 20 minutes,the tumor cell lines are detected at 525 nm by Flow CytoMeter.Results: Soybean flavones have obviously inhibition to liver cancer SMMC7721 cell in 24 h,48 h and 72 h,and the inhibiting rates increase following the flavones concentration raise.After treatment of the soybean flavones for 24 hours,the numbers of CD133 tumor stem cells have obvious decrease.Conclusion: Soybean flavones not only inhibit the proliferation of tumor cells,but also inhibit CD133 tumor stem cells.
