正交设计研究艾灸对溃疡性结肠炎大鼠穴区温度和Cx43表达的作用

摘    要:目的采用正交实验设计方法,观察麦粒灸三因素三水平不同灸量对溃疡性结肠炎(UC)大鼠穴区温度和Cx43表达的影响。方法 90只SD大鼠随机抽取8只作为正常组,余82只大鼠采用免疫学方法加局部刺激制备UC模型,在确认模型成功的基础上分为模型组与麦粒灸干预组,麦粒灸干预组又随机分为9个不同灸量干预亚组,以三因素(频次、疗程、壮数)三水平组成的灸量于UC大鼠双侧天枢穴施麦粒灸。结果与结论麦粒灸三因素三水平组合的不同灸量对UC大鼠灸后即刻、灸后5min穴区皮肤温度以及Cx43阳性表达具有不同的影响,对灸后10min穴区皮肤温度的影响无显著差异。艾灸壮数是影响灸后即刻穴区皮肤温度的主效应因素,2次/d、3壮/次、5d的灸量组合作用最显著;频次是影响灸后5min穴区皮肤温度的主效应因素;疗程是影响灸后穴区Cx43阳性表达的主效应因素,1次/2d、3壮/次、10d的灸量组合作用最显著。

Abstract:Objective To investigate,by an orthogonal test,the effect of three-factor and three-level different amounts of wheat-grain-sized moxa cone moxibustion on acupoint area temperature and Cx43 expression in ulcerative colitis (UC) rats. Methods Eight SD rats selected randomly from 90 constituted a normal group. A UC model was made by an immunological method plus local stimulation in the remaining 82 rats. After it was ascertained that the model was made successfully,the rats were allocated to a model group and a wheat-grain-sized moxa cone moxibustion intervention group. The latter was again randomly allocated to 9 subgroups that would received different amounts of moxibustion intervention. Different amounts of wheat-grain-sized moxa cone moxibustion composed of three factors (frequency,course treatment and number of cones) and three levels were performed on bilateral points Tianshu (ST 25) in different subgroups of UC rats,respectively. Results and conclusion Different amounts of wheat-grainsized moxa cone moxibustion composed of three factors and three levels had different effects on UC rat acupoint area temperature and Cx43 expression immediately and at 5 min after moxibustion. There was no significant difference in the effect on acupoint area temperature at 10 min after moxibustion. The number of moxa cones is the main factor influencing the immediate effect of moxibustion on acupoint area skin temperature. A combination of moxibustion 3 cones/once,twice/day and 5 days has the most marked effect. The frequency of moxibustion is the main factor influencing its effect on acupoint area skin temperature at 5 min after it. The course of treatment is the main factor influencing the effect of moxibustion on acupoint area Cx43 expression. A combination of moxibustion 3 cones/once,once/2 days and 10 days has the most marked effect.
