HYAL1基因过表达对乳腺癌细胞迁移能力和血管生成能力及增殖的影响

目的探讨透明质酸酶HYAL1基因过表达对人乳腺癌细胞株MCF-7和ZR-75-30的迁移能力和血管生成能力以及增殖的影响。方法应用双室联合培养技术,建立体外肿瘤侵袭和血管生成模型,观察过表达HYAL1基因对乳腺癌细胞穿透ECM gel和诱导血管内皮细胞形成血管能力的影响;应用MTT和流式细胞术检测细胞的增殖能力变化。结果过表达HYAL1基因的乳腺癌细胞MCF-7和ZR-75-30(实验组)穿透ECM的能力以及诱导血管生成的能力均强于对照组(P<0.05);但是,过表达HYAL1基因的乳腺癌细胞增殖能力与对照组比较没有明显差异(P>0.05)。结论过表达HYAL1基因能促进人乳腺癌细胞在体外侵袭和诱导血管生成,但是对乳腺癌细胞的增殖没有影响。
