黄芪对缺血再灌注心肌细胞凋亡的影响

摘    要:目的:探索黄芪对大鼠急性心肌炔血再灌注时心肌细胞凋亡及凋亡相关基因表达的影响。方法:采用结扎大鼠左冠状动脉法制备心肌缺血再灌注(IR)模型,分别以地高辛随机引物法及免疫组化法检测心肌凋亡细胞、心肌细胞bax、bcl—2基因的蛋白表达情况。结果:缺血再灌注心肌细胞凋亡数量显著高于假手术组(P<0.01),黄芪治疗组心肌细胞凋亡(P<0.05)明显受到抑制。IR组和黄芪组bax和bcl--2基因蛋白表达均明显增加(P均<0.01),黄芪明显抑制bax基因表达(P<0.05),但对bcl—2基因表达无明显影响。结论:急性心肌缺血再灌注时心肌细胞凋亡加剧,bax、bcl—2参与了缺血再灌注时心肌细胞凋亡凋控,黄芪可抑制凋亡加速基因bax的表达,因而在缺血再灌注损伤心肌细胞凋亡的保护方面有一定价值。

Abstract:Objective To investigate the effects of Huangqi on myocyte apoptosis and the expression of apoptosis-associated genes in rat hearts with acute myocardial ischemia and reperfusion.Methods Models were made by a ligation or release of left coronary artery in rats.Myocyte apoptosis and the protein expression of bax and bcl-2 genes were detected by digoxin random labeling and immunohistochemistry.Results The apoptotic index and the positive expression index of bax and bcl-2 genes were significantly increased in IR group.Huangqi significantly inhibitted the myocyte apoptosis and the expression of bax gene,but had no effect on bcl-2 gene expression.Conclusion The myocyte apoptosis and the expression of bax and bcl-2 are significantly increased in rat hearts with IR.Huangqi is valuable in the treatment of IR because of its inhibition to bax gene expression.
