摘    要:目的 研究高强度聚焦超声 (HIFU)及其联合甲氨喋呤 (MTX)对人膀胱癌细胞株T2 4的体外效应。方法 不同剂量HI FU单独及联合MTX作用T2 4细胞悬液后观察细胞存活率及生长增殖的变化。结果 一定剂量的HIFU可使T2 4细胞存活率直线下降并产生短暂的生长抑制作用 ;低剂量HIFU(11.5W/cm2 × 6秒 )可使MTX对T2 4细胞抑制率增加 ;HIFU(45 0W/cm2 × 6秒 )与MTX联合作用对T2 4细胞生长增殖的抑制作用明显强于单用HIFU及单用MTX。结论 在体外以空化、机械效应为主的HIFU对T2 4细胞具有急性致死及短暂的生长抑制作用 ;对MTX具有增敏作用 ;联合MTX作用具有协同效应。

Abstract:Objective To study the effects of high intensity focused ultrasound (HIFU)and its combined with MTX on human bladder cancer cell. Methods After action by different intensities HIFU/combined with MTX, observe the changes of T24 cell survival rate、growth curves. Results A certain intensity HIFU could decrease the cell survival rate and inhibit temporarily cell growth. Theirradiation of HIFU(11.5W/cm2×6s) could enhance MTX inhibition to I24 cell. HIFU ( 450W/cm2×6s) combined with MTX could halt T24 cell growth in 10d. It was stronger than HIFU alone and MTX alone. Conclusion In vitro edfects of HIFU which the main effection was cavitation and mechanical effection could kill T24 cell and inhibit temporarily cell growth. It could potentiate sensitivity of MTX. HIFU combined with MTX has synergistic inhibition to T24 cancer cell line.
