瘢痕灸对荷瘤及化疗小鼠肠道淋巴结及固有层淋巴细胞亚群的影响

摘    要:目的观察瘢痕灸对肿瘤化疗肠道黏膜损伤的治疗作用及其对黏膜免疫中肠道集合淋巴结(PP结)及固有层中淋巴细胞(LPL)亚群的影响。方法运用给小鼠原位接种C-26结肠癌并重复灌胃环磷酰胺,建立肿瘤化疗肠道黏膜损伤的模型,瘢痕灸处理17d,观察瘢痕灸对瘤重、抑瘤率及PP结面积的影响,分离肠PP结内的淋巴细胞,用免疫荧光染色及流式细胞仪分析黏膜免疫各部位T淋巴细胞亚群的变化。结果环磷酰胺组瘤重显著降低,抑瘤率约为37.59%;环磷酰胺加灸组瘤重与环磷酰胺组比较无统计学意义。荷瘤及环磷酰胺都可显著降低PP结面积,明显增加PP结内CD3^+和CD8^+细胞百分比,降低LPL中CD4^+细胞百分比。而瘢痕灸可明显减轻环磷酰胺对PP结面积的影响(P〈0.01)。瘢痕灸还能使荷瘤和环磷酰胺作用下显著减少LPL中的CD。’细胞百分比显著增加(P〈0.05),但对LPL中的CD^8+细胞百分比及PP结中的CD3^+、CD4^+、CD8^+T细胞亚群无明显影响。结论荷瘤及环磷酰胺可明显抑制肠道黏膜免疫功能,而瘢痕灸能减轻荷瘤及环磷酰胺对PP结及LPL中某些T细胞亚群的影响,改善黏膜免疫抑制状态。

Effect of Scarring-moxibustion on Peyer's Patch and T Cells Subsets of Intraepithelial Lymphocyte of Tumor-bearing Mice with cyclophosphamide

Abstract:Objective To explore the effect of scarring-moxibustion on Peyer's patch and T cells subsets of intraepithelial lymphocyte of tumor-bearing mice with cyclophosphamide. Method The mucosal immune deficiency model was made by in site inoculating C-26 colon carcinoma with repeated intragastric administration of cyclophosphamide. The mice were treated with scarringmoxibustion for seventeen days after inoculation. The area of Peyer's patch were measured, and T cells subsets of Lamina propria lymphocyte (LPL) were separated and detected by immunofluorescence test and flow cytometer. Result Tumor weight of tumor-bearing group was higher than cyclophosphamide group significantly (P〈0.01). The tumor inhibition ratio of cyclophosphamide was 37.59%. The area of Peyer's patch of intestine in tumor-bearing group decreased significantly.The amounts of CD4^+ cell of LPL was decreased in the tumor-bearing group and cyclophosphamide group. Scarring-moxibustion could significantly increase the area of PP and the amounts of CD4^+ cell of LPL (P〈0.01, P〈0.05), while had no effect on CD8^+ cell of LPL and T ceils subsets in Peyer's patch.Conclusion Scarring-moxibustion can improve the mucosal immune defective caused by tumor and cyclophosphamide.
