结肠癌组织中SATB1的表达及其临床意义

目的探讨SATB1异常表达与结肠癌发生、发展及预后的关系。方法分别用RT-PCR和Western blot法检测SATB1在25例结肠癌活检组织及相应癌旁正常组织中mRNA和蛋白表达水平;采用免疫组化检测58例结肠癌石蜡组织切片中SATB1的表达,分析其与结肠癌临床病理因素的关系及预后意义。结果 SATB1 mRNA和蛋白在92%(23/25)结肠癌活检组织中的表达水平高于其癌旁正常组织;58例结肠癌石蜡组织中,SATB1蛋白强阳性率为46.6%(27/58)。统计学分析结果显示,SATB1强阳性与结肠癌Dukes分期和远处转移密切相关,而与性别、年龄、病理分级、肿瘤大小、淋巴结转移无显著相关性。结论 SATB1在结肠癌中高表达,其表达水平与肿瘤进展相关,可作为预测结肠癌进展和转移的分子标记物。

Expression of SATB1 and its clinicopathologic significance in colorectal cancer

Objective To investigate the expression level and clinicopathologic significance of SATB1 in colorectal cancer.Methods SATB1 mRNA and protein expression in 25 pairs of colorectal cancer(CRC) biopsies and their matched adjacent tissues were detected by RT-PCR and Western blot,respectively.Immunohistochemical analysis was used to examine the expression of SATB1 in 58 cases of paraffin-embedded colorectal cancer tissues.Results Both of SATB1 mRNA and protein expression levels were significantly higher in 92%(23/25)colorectal cancer biopsies than in their matched adjacent tissues.SATB1 was strongly expressed in 46.6%(27/58) paraffin-embedded colorectal cancer tissues.Additionally,strong expression of SATB1 was significantly associated with Dukes stage and distant metastasis,while not with gender,age,pathologic stage,tumor size and lymph node involvement.Conclusions High expression of SATB1 is closely related to progression of CRC.SATB1 may serve as a new molecular marker for prediction of colorectal cancer progression and metastasis.
