多发性硬化症患者外周血辅助性T细胞17、调节性T细胞及其相关炎症因子和血清铁蛋白水平变化

摘    要:目的 观察多发性硬化症(MS)患者外周血辅助性T细胞17(Th17细胞)、调节性T细胞(Tr细胞)及血清铁蛋白、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平的变化,并探讨其在MS发生发展中的临床意义。方法 选择2018年1月至2021年12月新乡医学院第一附属医院收治的46例MS患者为MS组,另选择年龄、性别及体质量指数相匹配的46名健康者为对照组。采用流式细胞术检测2组受试者外周血Th17细胞和Tr细胞水平,并计算Th17细胞与Tr细胞的比值(Th17/Tr);磁微粒化学发光法检测血清TNF-α、IL-6水平,免疫比浊法检测血清铁蛋白水平。结果 MS组患者外周血Th17细胞水平及Th17/Tr显著高于对照组,Tr细胞水平显著低于对照组(P<0.05);复发进展期MS患者外周血Th17细胞水平及Th17/Tr显著高于缓解期患者,Tr细胞水平显著低于缓解期患者(P<0.05);缓解期MS患者外周血Th17细胞、Tr细胞水平及Th17/Tr与对照组受试者比较差异无统计学意义(P>0.05)。MS组患者血清TNF-α、IL-6水平显著高于对照组(P<...

Abstract:Objective To observe the changes of peripheral blood helper T cell 17 (Th17 cell),regulatory T cell (Tr cell) and serum ferritin,tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6) levels in patients with multiple sclerosis (MS),and to investigate their clinical significance in the development of MS.Methods Forty-six MS patients admitted to the First Affiliated Hospital of Xinxiang Medical University from January 2018 to December 2021 were selected as the MS group,and 46 healthy persons matched in age,sex and body mass index were selected as the control group.The levels of Th17 cells and Tr cells in peripheral blood of subjects in the two groups were detected by flow cytometry,and the ratio of Th17 cells to Tr cells (Th17/Tr) was calculated.The levels of serum TNF-α and IL-6 were detected by magnetic particle chemiluminescence method,and the level of serum ferritin was detected by immunoturbidimetry.Results The levels of peripheral blood Th17 cells and Th17/Tr in the MS group were significantly higher than those in the control group,while the level of Tr cells was significantly lower than that in the control group (P<0.05).Compared with MS patients in remission stage,the levels of Th17 cells and Th17/Tr in peripheral blood of MS patients in relapse progression stage were significantly increased,while the level of Tr cells was significantly decreased (P<0.05).There was no significant difference in the levels of peripheral blood Th17,Tr cells and Th17/Tr between the MS patients in remission stage and the subjects in the control group (P>0.05).The levels of serum TNF-α and IL-6 of patients in the MS group were significantly higher than those in the control group (P<0.05),while there was no significant difference in serum ferritin level between the two groups (P>0.05).The levels of serum TNF-α and IL-6 in MS patients in relapse progression stage were significantly higher than those in patients in remission stage (P<0.05).There was no significant difference in serum ferritin level between MS patients in relapse progression stage and remission stage (P>0.05).There was no significant difference in serum TNF-α,IL-6 and ferritin levels between the patients with MS in remission stage and the subjects in the control group (P>0.05).The Th17/Tr in peripheral blood was positively correlated with the levels of serum TNF-α and IL-6 in MS patients (P<0.05),while there was no significant correlation between the levels of Th17 cells and Tr cells in peripheral blood and the levels of serum TNF-α and IL-6 in MS patients (P>0.05).Conclusion The Th17/Tr and the levels of related inflammatory factors TNF-α and IL-6 increased in MS patients,which may be related to the occurrence and development of MS.
