摘    要:
目的 探讨lncRNA H19(H19)在心力衰竭(heart failure, HF)中的临床意义以及通过转化生长因子-β(transforming growth factor-β,TGF-β)/重组人Smad3蛋白(SMAD family member 3,Smad3)信号通路对心肌细胞的影响机制。方法 选择2018年5月至2021年5月收治的327例HF患者(研究组)以及50例性别与年龄匹配的单纯糖尿病(或高血压)患者(对照组),分析H19在HF中的表达以及其临床意义。分离1~2日龄Sprague-Dawley大鼠心室肌细胞,构建H19低表达载体(si-H19)和TGF-β1过表达载体(pcDNA3.1),探究H19对心肌细胞凋亡的影响以及作用机制。结果 在HF患者中H19也出现表达水平上升,并且对HF具有一定诊断价值,其曲线下面积(area under curve, AUC)为0.874,并且H19是HF的独立危险因素。沉默H19后,TGF-β1/Smad3通路表达与心肌细胞凋亡均降低。值得注意的是,过表达TGF-β1仅部分恢复了沉默H19对心肌细胞凋亡的抑制作用。结论 H19...

Abstract:
Objective To investigate the clinical significance of lncRNA H19 (H19) in heart failure (HF) and its effect on cardiomyocytes through the transforming growth factor-β (TGF-β)/MAD family member 3 (Smad3) signaling pathway. Methods From May 2018 to May 2021, 327 HF patients were selected as the research group, as well as 50 sex- and age-matched patients with simple diabetes (or hypertension) were enrolled as control group, to analyze the expression of H19 in HF and its clinical significance. The 1-2 days old Sprague-Dawley rat ventricular myocytes were isolated. H19 low expression vector (si-H19) and TGF-β1 overexpression vector (pcDNA3.1) were constructed to investigate the effect of H19 on cardiomyocyte apoptosis and its mechanism. Results The expression level of H19 was increased in HF patients, and had a certain diagnostic value for HF. Its area under curve (AUC) was 0.874, and H19 was an independent risk factor for HF. After silencing H19, both the expression of TGF-β1/Smad3 pathway and cardiomyocyte apoptosis were sreduced. Notably, overexpression of TGF-β1 only partially restored the inhibitory effect of silencing H19 on cardiomyocyte apoptosis. Conclusion H19 is highly expressed in HF, promotes the apoptosis of cardiomyocytes by activating the TGF-β1/Smad3 signaling pathway, and participates in the development of HF.
