麦冬多糖调控胰高血糖素样肽-1对妊娠期糖尿病大鼠肠道菌群及糖代谢的影响

摘    要:目的:探究麦冬多糖调控胰高血糖素样肽-1(GLP-1)对妊娠期糖尿病大鼠肠道菌群及糖代谢的影响。方法:将80只孕鼠随机分为空白组、模型组、低剂量组、中剂量组、高剂量组、阳性对照组、Exendin-4组和高剂量+Exendin-4组,每组10只。干预2周后比较各组大鼠空腹血糖(FBG)和胰岛素抵抗指数(HOMA-IR)水平;ELISA法检测血清GLP-1水平;对各组大鼠进行16S-rDNA肠道菌群测序;RT-PCR法检测大鼠肝脏组织葡萄糖激酶(GCK) mRNA及脂肪组织葡萄糖转运体(GLUT4) mRNA表达。结果:与空白组比较,模型组大鼠FBG、HOMA-IR、拟杆菌门丰度、普氏菌属丰度、瘤胃球菌属丰度明显升高,血清GCK、GLUT4 mRNA及GLP-1表达水平、厚壁菌门丰度、厚壁菌门/拟杆菌门比值、乳酸杆菌属丰度、密螺旋体属丰度明显降低(均P<0.05);与模型组比较,低剂量组、中剂量组、高剂量组、阳性对照组和Exendin-4组大鼠FBG、HOMA-IR、拟杆菌门丰度、普氏菌属丰度、瘤胃球菌属丰度明显降低,血清GCK、GLUT4 mRNA及GLP-1表达水平,厚壁菌门丰度...

Abstract:Objective:To investigate the effects of ophiopogon polysaccharides on intestinal flora and glucose metabolism in gestational diabetic rats by regulating GLP-1.Methods:Eighty pregnant rats were randomly divided into blank group,model group,low-dose group,medium-dose group,high-dose group,positive control group,Exendin-4 group and high dose + Exendin-4 group,10 rats in each group.After 2 weeks of intervention,FBG and HOMA-IR were compared among all groups.Serum GLP-1 levels were determined by ELISA.16S-rDNA intestinal flora was sequenced in each group.the expression of GCK and GLUT4 mRNA was detected by RT-PCR.Results:Compared with the blank group,the FBG,HOMA-IR,the abundance of Bacteroidets,Prevotella,Ruminococcus were significantly higher,and the serum GLP-1 level,GCK,GLUT4 mRNA expression,Firmicutes/Bacteroidetes ratio,the abundance of Firmicutes,Lactobacillus,Treponema were significantly lower in the model group(all P<0.05).Compared with the model group,FBG,HOMA-IR,the abundance of Bacteroidets,Prevotella,Ruminococcus were significantly lower in the low-dose group,medium-dose group,high-dose group,positive control group and Exendin-4 group,and serum GLP-1 level,GCK,GLUT4 mRNA expression,Firmicutes/Bacteroidetes ratio,the abundance of Firmicutes,Lactobacillus,Treponema were significantly higher(all P<0.05).Compared with the high-dose group,FBG,HOMA-IR,the abundance of Bacteroidets,Prevotella,Ruminococcus were significantly lower in the high-dose + Exendin-4 group,and the serum levels of GLP-1,GCK,GLUT4 mRNA expression,Firmicutes/Bacteroidetes ratio,the abundance of Firmicutes,Lactobacillus,Treponema were significantly higher(all P<0.05).Conclusion:Ophiopogon polysaccharide could improve the dysregulation of intestinal flora in gestational diabetic rats by regulating GLP-1 levels,and also had a significant improvement on glucose metabolism.
