蛋白多糖在黏膜免疫模型鼠肠系膜淋巴结高内皮微静脉中的表达

摘    要:目的研究伤寒杆菌诱导的肠道黏膜免疫模型小鼠肠系膜淋巴结(MLN)内高内皮微静脉(HEVs)蛋白多糖(PG)的表达,探讨其对黏膜免疫淋巴细胞归巢的调控作用。方法采用阳性胶体铁染色光镜和透射电镜观察。结果蛋白多糖在伤寒杆菌诱导的黏膜免疫模型小鼠肠系膜淋巴结高内皮微静脉内皮细胞管腔面细胞膜呈强阳性表达。电镜下可见高内皮微静脉管腔内的淋巴细胞体积大小不等,多数细胞胞体较大,胞浆增多,细胞器丰富,淋巴细胞多呈活化状态。此外,还可见到多个淋巴细胞位于内皮细胞内、内皮细胞连接的间隙内以及基膜内,呈穿越状态。结论蛋白多糖对淋巴细胞穿越高内皮微静脉管壁以及向淋巴组织迁移可能有重要的调节作用。

Abstract:Objective To investigate the expression of proteoglycan in high endothelial venues(HEVs) of mesenteric lymph node(MLN) in mouse model of muscosal immunity to explore its possible role for lymphocyte homing in muscosal immunity. Methods The lymphocyte morphology and proteoglycan expression in HEVs of MLN in mouse model of muscosal immunity inducted by salmonella typhi were observed by light microscopy with cationic iron colloid solution staining and transmission electron microscopy. Results The expression of proteoglycan in HEVs lumens of MLN showed strong positive. Lymphocytes showed different in volume with more plasma and organelles in HEVs lumens, some were plasmocyted with plenty reticulum in cytoplasm, some membrane connected with endotheliocyte and in active status. Conclusion The overexpression of proteoglycan in HEVs of MLN might be related to the pathogenesis of muscosal immunity.
