| 摘 要： | 目的：研究常用化疗药物对SGC－7901人胃癌细胞株端粒酶活性的调节作用，探讨端料酶作为化疗敏感性指标的可行性。方法：选择顺铂、丝裂霉素C、阿霉素及5－氟尿嘧啶和甲酰四氢叶酸处理胃癌细胞，采用半定量TRAP－银染法检测药物处理前后的端粒酶活性，流式细胞学检测细胞周期和细胞凋亡。结果：顺铂、丝裂霉素C和阿霉素在诱导SGC－7901细胞凋亡的同时，下调端粒酶活性，且抑制作用呈时间依赖关系，5－氟尿嘧啶和甲酰四氢叶酸对端粒酶活性无明显抑制作用。结论：观测细胞株端粒酶的活性变化可作为化疗药物敏感性的指标，可合理选择新的化疗药物。

| Abstract: | Objective:To investigate the modulation on telomerase activity of gastric cancer cell line by several drugs that often used in gastric cancer treatment and to determine the chemosensitive value of telomerase by semi TRAP assay.Methods:Telomerase activities of SGC 7901 cell line treated by chemotherapeutic drugs(cisplatin,mitomycin c,doxorubicin,or 5 fluorouracil and folinate)were detected by semi TRAP assay.Also FCM was used to determine cell cycle and apoptosis at the same time.Results:Chemotherapeutic drugs cisplatin,mitomycin c and doxorubicin inhibited telomerase activity in a time dependent manner.FCM results showed apoptosis was detected before the changes of telomerase activity,while 5 fluorouracil and folinate showed no significant inhibition on telomerase.Conclusion:Detection of telomerase activity by semi TRAP assay after treatment with chemotherapeutic drugs could be used as a chemosensitivity target. |
