| 摘 要： | 目的 探讨吸烟对入骨组织骨桥蛋白(OPN)表达及功能的影响.方法 随机选取吸烟和不吸烟的胫骨骨折健康成年志愿者各10名,分为吸烟组和非吸烟组.吸烟者每天吸烟20支以上,烟龄不少于10年;非吸烟者每天吸烟不超过1支或不吸烟.在胫骨骨折手术中取约1克胫骨近端干骺端松质骨,提取出总RNA和总蛋白,通过逆转录聚合酶链反应(RT-PCR)测定OPN mRNA的表达,通过蛋白印迹(Western blotting)测定OPN的表达,通过免疫沉淀法测定OPN的磷酸化程度.结果 吸烟组RT-PCR条带光密度值的OPN/β-肌动蛋白(β-actin)比值高于非吸烟组,差异有显著性(P<0.05).吸烟组OPN蛋白印迹检测的条带光密度值高于非吸烟组(P<0.01).吸烟组OPN免疫沉淀检测的条带光密度值高于非吸烟组(P<0.01).结论 吸烟可导致入骨组织OPN mRNA与OPN表达增加,OPN磷酸化程度增强.
