摘    要:目的建立一种可快速测定多种临床常用喹诺酮类药物浓度的PR-HPLC法。方法以Scienhome,C18不锈钢柱(250 mm×4.6 mm,5μm色谱柱),0.01 mol/L磷酸二氢钾-甲醇-0.5 mol/L四丁基溴化氨(78∶22∶4,V/V)为流动相,流速:1.0 mL/min。柱温:40℃,波长依受检药物为定,灵敏度:0.001 AFU。外标法定量。以二氯甲烷(含2%异丙醇)萃取样品,氮气吹干后以流动相复溶,进样20μL。结果按上述色谱条件和样品处理,左氧氟沙星、莫西沙星等5种喹诺酮类药物可得到可靠分离和准确检测。结论所建立的PR-HPLC法简便易行,准确可靠。可在基层医院对多种喹诺酮类药物进行血药浓度检测。

Abstract:Objective To develop a PR-HPLC method for the fast simultaneous determination of various quinolone antimicrobial agents in human plasms. Methods The separation was conducted on the Seienhome, C18 column was used (250 mm×4.6 mm,5μm column). The mobile phase consised of 0.01 mol/L KH2PO4 buffer solution-Methanol- 0.5 mol/L Tetrabutyl ammonium bromide with a volume ratio of 78:2:4(V/V). The flow rate was 1.0 mL/min. The determined wavelength depended on the determined quinolone antibiotics. The column temperrature was 40℃ and the injection volume was 20 μL. Liquid-liquid extraction with Methylene chloride (containing 2% Dimethylearbinol) was screened for the sample processing. After dried under nitrogen, the residue was redissolved with mobile phase and injected into the sampler. Results Under the optimized chromatographic conditions various quinolone antimicrobial agens, such as Levofloxacin Hydrochloride and moxifloxacin could be separated effectively and detected exatly. Conclusion The established PR-HPLC method is accurate, simple and rapid, which can be used for the simultaneous determination of various quinolone antimicrobial agents in basic level hospital.
