氧化铁磁流体热疗对人良性脑膜瘤细胞增殖和凋亡的影响

摘    要:目的观察氧化铁磁流体热疗(MFH)对人良性脑膜瘤细胞增殖和凋亡的影响。方法15例人脑膜瘤细胞短期原代培养,进行氧化铁磁流体单次加热或3次加热,加热温度42~44℃,持续30min。流式细胞仪检测细胞凋亡,四氮唑蓝(MTT)比色法检测细胞增殖的变化。结果MFH(44℃,30min)单次加热和3次加热组中,人脑膜瘤细胞增殖抑制率分别为(12.62±0.78)%和(12.78±1.56)%,与对照组相比差异显著(P0.05)。MFH引起部分脑膜瘤细胞裂解坏死,散在细胞凋亡,但各组细胞凋亡率无明显差异。结论在人良性脑膜瘤体外实验中,氧化铁MFH(44℃,30min)可以明显抑制细胞增殖,但不能有效诱发细胞凋亡。

Abstract:Objective To investigate the effect of magnetic fluid hyperthermia (MFH) on the proliferation and apoptosis of human benign meningiomas. Methods The short-term primary culture of the human fresh meningioma cells derived from 15 patients was performed. The third generation cells were once or thrice heated to 42~44 ℃ for 30 minutes by ferric oxide MFH. The cells apoptosis was detected by flow cytometry. The inhibitory rate of the meningiomas cells proliferation was determined by methyl thiazolyl tetrazolium (MTT) assay 1, 2, 3 and 4 days after the treatment with MFH. Results The inhibitory rates of meningioma cells proliferation in once heating (44 ℃, 30 min) group and thrice heating (44 ℃, 30 min) group were (12.62±0.78)% and (12.78±1.56)% respectively 4 days after heating with MFH, which were significantly higher than that in the control group (P0.05). There mainly was timorous cells necrosis in all the heating groups, in which the rates of the cell apotosis were insignificantly different from that in the control group. Conclusion In vitro, the proliferation of human meningiomas cells can be inhibited by heating to 44 ℃ for 30 min with ferric oxide MFH, by which the tumor cell necrosis, not apoptosis can be induced.
