水蛭抗凝血活性成分的研究

摘    要:目的:研究水蛭的抗凝血成分。方法:以血浆复钙时间为指标,应用Sephadex DEAE A-50阴离子交换树脂色谱、Sephadex G-25和Sephadex LH-20凝胶过滤色谱及反相高压液相色谱技术分离纯化。结果:得到3个抗凝血活性多肽,其中化合物1,2在自然条件下互相转化,它们的相对分子质量分别为7 100和5 531;采用HPLC及SDS-PAGE鉴定了化合物3的纯度,采用MALDI-TOF-MS法测定出其相对分子质量为8 608,并测定了其氨基酸组成。结论:推测化合物3为首次分离得到的抗凝血多肽,命名为蚂蟥多肽(whitmanin)。

Abstract:Objective: To study the anticoagulant constituents in dried leech (Whitmania pigra Whitman). Method: The plasma recalcification time (PRT) as index, the constituents with anticoagulant activity were isolated and purified by anion-exchange chromatography on Sephadex DEAE A-50, gel permeation chromatography on Sephadex G-25 and Sephadex LH-20 columns, and then reversed phase high-performance liquid chromatography successively. Result: Three anticoagulant polypeptides were isolated and purified. Compounds 1 and 2 can be translated each other in natural conditions, and their molecular weights are 7 100 and 5 531, respectively. Compound 3 was identified as a pure polypeptide by HPLC and SDS-PAGE, and its molecular weight was determined as 8 608 by MALDI-TOF-MS. The amino acid composition of compound 3 was also determined. Conclusion: Compound 3 was inferred to be a novel anticoagulant, and named whitmanin.
