氟对体外培养大鼠成骨细胞钙调蛋白表达影响

摘    要:目的 观察不同浓度的氟在不同时间段对体外培养大鼠成骨细胞的钙调蛋白(CaM)的影响。方法 采用骨组织块法分离培养新生Wistar大鼠颅骨成骨细胞,将不同浓度的氟作用于大鼠成骨细胞,分别于染氟培养24,36和48h采用免疫细胞化学法检测CaM蛋白的表达。结果 在染氟培养24h,与对照组比较,0.5mg/L氟组成骨细胞CaM蛋白的表达明显增加,2mg/L氟组CaM蛋白的表达明显减少;染氟培养36h,与对照组比较,2mg/L氟组CaM蛋白表达明显增加,8mg/L氟组CaM蛋白的表达明显减少;而在染氟培养48h,各氟组CaM蛋白的表达较对照组均显著增加。结论 氟在一定浓度、时间范围内对成骨细胞的CaM蛋白表达影响的表现为先抑制后促进的作用。

Abstract:Objective To study the effects of fluoride on expression of calmodulin(CaM)in rat osteoblasts cultured in vitro in different doses and at different time.Methods Osteoblasts were isolated from the calvariae of new-born Wistar rats by means of bone tissue block,and cultured with DMEM.Different concentrations of fluoride were added to rat osteoblasts and incubated for differenthours(24 hours,36 hours and 48 hours).The expression of CaM was analyzed with immunohisto-chemical stain.Results At 24 hours,0.5 mg/L fluoride significantly increased the expression of CaM in rat osteoblasts,but 2 mg/L fluoride decreased it.At 36 hours,the expression of CaM was higher in 2 mg/L fluoride group and lower in 8 mg/L fluoride group than that in control group(P<0.05).At 48 hours,the expression of CaM in all fluoride groups were significanthigher than that in control group(P<0.05).Conclusion The fluoride effect on the expression activity of CaM in osteoblasts is dose-and time-dependent and the effect of inhibiting is ahead of the effect of stimulating.
