神经元移植与鼠颞叶癫痫海马γ-氨基丁酸能神经元的变化

摘    要:目的探讨脂肪干细胞(ADSCs)转化神经元移植前后大鼠颞叶癫痫海马γ-氨基丁酸(GABA)能神经元的变化及海马形态学改变。方法40只SD大鼠随机分为3组,移植组(n=15):海马内注射海人酸(KA)后,依据大鼠左侧海马CA3区坐标,立体定向将ADSCs转化神经元注入海马;移植对照组(n=15):海马内注射KA后,靶点内移植生理盐水;生理盐水对照组(n=10):海马内注射生理盐水后,不进行任何移植。比较手术前后大鼠海马GABA能神经元的变化以及海马形态学的改变。结果ADSCs转化神经元移植后,GABA能神经元较移植前显著增加(P<0.01)。移植对照组海马区神经元水肿数量较生理盐水对照组明显增多,细胞内胞质贫乏,细胞器数量减少,细胞核边界不规整;而移植组神经元水肿减轻,其余结构变化与生理盐水对照组差异不大;且术后8周较2周与生理盐水对照组的差异更小。结论ADSCs转化神经元移植对颞叶癫痫大鼠的神经元修复起到了良好的效果。

Abstract:Objective To explore the the changes of hippocampus gamma-aminobutyric acid(GABA) neurons and hippocampus morphology in the temporal lobe epilepsy rat models before and after transplantation of neural cells from adipose tissue-derived stem cells(ADSCs). Methods Forty SD rats were randomized into three groups.Transplantation group(n=15): after injection with kainic acid(KA) into the hippocampus,neural cells from ADSCs were slowly injected into CA3 area of the left hippocampus according to the coordinate and stereotactic management.Transplantation control group(n=15): after injection with KA into the hippocampus,normal sodium was transplanted.Normal sodium group(n=10): only normal sodium was injected,with no transplantation.The changes of hippocampus GABA neurons and hippocampus morphology were compared before and after the operation. Results After transplantation,the GABA neurons of transplantation group were significantly more than transplantation control group(P<0.01).The hippocampus neurons in transplantation control group were found edema and were more severe than normal sodium group,cytoplasm and organelle were reduced and the karyon was not complete.However,the edema of rats hippocampus neurons in transplantation group was improved.The cell structure was similar to that of normal sodium group,and this was more obvious for the 8th week than the 2nd week. Conclusion The transplantation of neural cells from ADSCs may play a part in the repair of temporal lobe epilepsy rat neurons.
