| 摘 要： | 目的淫羊藿苷对体外培养的人外周血内皮祖细胞(EPCs)增殖的影响。方法采用密度梯度离心法分离获得人外周血单个核细胞,用添加VEGF、FGF的M199培养液培养7d后,经激光共聚焦显微镜、流式细胞仪鉴定为EPCs。用不同浓度药物干预培养EPCs,分别作用不同时间(24、48、72h)后,倒置相差显微镜下观察EPCs生长情况,四唑蓝比色试验(MTT法)测定EPCs的吸光度值。用西药西辛伐他汀作为对照组。结果与对照组比较,淫羊藿苷高、中、低浓度组均改善了EPCs增殖功能(P<0.05),且呈一定的量效、时效关系,在中浓度最为显著(P<0.05)能明显促进增殖能力;在同一浓度组中,随时间延长淫羊藿苷对EPCs的促进作用逐渐增强,于72h达到高峰,且中药中剂量组对EPCs的促进作用优于西药组辛伐他汀组。结论淫羊藿苷能促进EPCs增殖,从而刺激缺血组织血管新生和再生,可能是其防治心脑血管及周围血管疾病的机制之一。 |
