摘    要:目的:观察CD81在正常大鼠视网膜色素上皮层(retinal pigment epithelium,RPE)的表达。方法:用抗CD81抗体EAT2对正常大鼠RPE组织切片及体外培养RPE进行免疫组织化学染色,H121对RPE组织进行CD81蛋白的Western blot分析。结果:正常大鼠视网膜的色素上皮细胞膜表面呈CD81的阳性染色,体外培养的大鼠RPE也呈CD81阳性表达;Western blot分析RPE层蛋白出现CD81阳性条带。结论:正常大鼠的RPE层可以表达CD81。

Abstract:AIM:As a member of tetraspanins superfamily,CD81 is proved to be involved in the inhibition of cellular mitotic activity,including cells of retinal pigment epithelium(RPE).The present study is to examine the distribution of CD81 in RPE of normal rats.METHODS:The distribution of CD81 was examined through EAT2 immunohistochemical staining on RPE of normal rats both in vivo and in vitro.A Western Blot analysis was performed to define the expression of CD81 protein in the RPE of rats.RESULTS:CD81 was found to express in the RPE of normal rats in vivo.The CD81 positive staining existed also on the surface of RPE in vitro and concentrated at regions of cell-cell contact.By Western Blot analysis,the expression of CD81 in the RPE was further confirmed.CONCLUSION:CD81 could be expressed in the RPE of normal rats.
