小鼠胎肝造血基质细胞株的建立及生物学特性和功能研究

摘    要:小鼠胎肝造血基质细胞经原代和传代培养并克隆成细胞株。该细胞株单层贴壁生长,核分裂相较多,有丝分裂指数较高,生长有赖于马血清的存在,有密度依赖抑制现象,染色体众数4n近80者常见。在半固体培养中无集落形成,ConA凝集试验阴性。该细胞株能双向调控红系和粒系造血,其刺激性影响主要由体液因子介导,其抑制性作用尚含细胞间短距离的调节因素。其培养上清液包括无血清培养上清液具有支持红系和粒系造血祖细胞增殖分化的活性,文中讨论了实验结果与胎肝临床疗效间的可能关系。

Abstract:A hemopoietic stroma cell strain from mouse fetal liver (MFLSC) has been established through a serial primary passage and cloned culture. This cell strain grew adherently and monolayerly, it had numerous nuclear mitotic features and a high index of mitosis. It's growth was dependent upon the presence of 15% or more horse serum with a density-dependent inhibition. The chromosome numbers of most of these cells were near 80. There was no colony formed by MFLSC in the semisolid culture system. MFLSC showed a negative aggregation reaction with ConA.The mouse fetal liver stromal cell strain showed both stimulating and inhibitory influence on the growth of erythropoietic and granulocyte-macrophage progenitor cells. The stimulating effect was mainly mediated by humoral factors, while the inhibitory influence could also be dependent upon the short-range contact regulation.The supernatant of MFLSC could be used in laboratory as exogenous sources of erythropoietic and granulopoietic stimulating factors instead of commonly applied EPO, BPA and GM-CSF.The possible relationship between hemopoietic stroma cells and the effect of fetal liver transfusion therapy in clinic was discussed.
