Cyclins/DNA多参数流式细胞术分析细胞增殖

摘    要:应用Cyclins/DNA双参数流式细胞术,分析有丝分裂原刺激后的正常人外周血T淋巴细胞的细胞增殖(特别是G1期细胞增殖),同时与Ki-67/DNA双参数法进行比较。结果表明,Cyclins/DNA法比Ki-67/DNA法早20h发现增殖的淋巴细胞,有丝分裂原刺激后48h,Cyclins/DNA法又比Ki-67/DNA法多检出20%的增殖淋巴细胞。结论:Cyclins/DNA双参数流式细胞术分析细胞增殖具可行性、实用性、科学性和全面性。

Abstract:The mitogen stimulated normal human T lymphocytes proliferation (especially G1 phase proliferation) was analyzed by using a developed cyclins/DNA multiparameter flow cytometry and Ki67/DNA multiparameter method. The results showed that the developed method detected proliferated T lymphocytes 20 earlier than Ki67 method, and 20 % more proliferated cells than Ki67 method at 48 h after mitogen stimulation. It was concluded that cyclins/DNA multiparameter folw cytometry is more practicable, reasonable and easy for cell proliferation analysis in contrast to the previous methods, especially for cell proliferation in early G1
