高效液相色谱法测定愈肠颗粒中生物碱的含量

目的研究建立愈肠颗粒中功劳木药材中小檗碱型生物碱高效液相色谱测定方法。方法采用高效液相色谱法,色谱柱：资生堂CAPCELL PAK C18（250mm×4.6mm,5μm）;流动相：乙腈-0.05mol/L磷酸二氢钠溶液（用磷酸调节pH值至3）（28∶72）;流速：1.0ml/min;检测波长：265nm;柱温：室温。结果盐酸巴马汀、盐酸药根碱、盐酸小檗碱分别在0.0354-0.354μg、0.0382-0.382μg、0.0207-0.207μg范围内,浓度与峰面积线性关系良好（r=0.9997,r=0.9998,r=0.9999）。平均回收率分别为：99.8%,100.3%,98.8%,RSD分别为1.7%、1.3%、1.8%。结论本方法简便,可作为愈肠颗粒的含量测定方法 。

High Performance Liquid Chromatography by Determination of Protoberberine in Yuchang Keli

Objective To establish a method for the determination of protoberberine in Compound Yuchang Keli by high performance liquid chromatography.Methods The high performance liquid chromatography system consisted of CAPCELL PAK C 18 column（250 mm×4.6 mm,5 μm）;with acetonitrile-0.05 mol/L NaH2PO4（pH adjusted to 3 with H3PO4）（28∶ 72）as the mobile phase.That flow rate was 1.0 ml/min and detected with UV at 265 nm.Results A good linear relationship was found within the range of 0.0354-0.354 μg of palmatine hydrochloride,0.0382-0.382 μg of tatrorrhizine hydrochloride and 0.0207-0.207 μg of berberine hydrochloride.The correlation coefficient（r）was 0.9997,0.9998 and 0.9999 respectively.The result（n=6）of recovery was 99.8%（RSD=1.7%）,100.3%（RSD=1.3%）and 98.8%（RSD=1.8%）.Conclusion The method is simple and suitable for determination of preparations.
