SOX2在肝细胞肝癌组织中的表达及其临床意义

目的：研究SOX2在肝细胞肝癌和癌旁肝组织的表达及其意义，探讨其表达水平与临床病理因素的联系。方法：收集42例肝细胞肝癌患者的临床资料，采用RT—PCR和Westernblot方法检测SOX2在42例肝细胞肝癌和癌旁肝组织的表达，并分析其表达水平与临床病理因素的联系。结果：PCR结果显示SOX2mRNA在肝细胞肝癌组织中的表达水平为0．7136±0．1556，明显高于癌旁肝组织（O4429±0．1215），差异有统计学意义（f=8．8864．P〈0．001）；Westernblot结果显示，SOX2蛋白在肝细胞肝癌的表达水平为0．4638±0．1600，高于癌旁肝组织（0．3130±0．1554），差异有统计学意义（t=3．8839，P〈0．05）；SOX2mRNA表达水平与患者的性别、年龄、血清AFP水平、肿瘤大小无明显相关性（P〉O．05），而与临床TNM分期有明显相关性（P〈0．05）。Westernblot显示SOX2蛋白在TNMI期中的表达明显低于Ⅱ、Ⅲ期中的表达。结论：SOX2在肝细胞肝癌组织中的表达高于癌旁肝组织中的表达，其表达水平与临床TMN分期密切有关。

Expression and significance of SOX2 in hepatocellular carcinoma

Objective: To study the impression and significance of SOX2 in hepatocellular carcinoma(HCC)and adjacent liver tissue.Methods: RT-PCR and Western Blot was used to determine the expression of SOX2 in HCC and adjacent liver tissue of 42 patients.The correlation between the SOX2 expression and clinical and pathological feature was statisticall analyzed.Results: RT-PCR showed that in 42 patients with hepatocellular carcinoma and adjacent liver tissue samples,SOX2 mRNA expression level in the liver with disabilities0.713 6±0.155 6,significantly higher than the adjacent liver tissue(0.442 9±0.121 5),the difference was statistically significant(t=8.886 4,P<0.001);Western Blot results,SOX2 protein expression level in the liver with disabilities 0.160 0 0.4638,its expression is higher than the adjacent liver tissue(0.313 0±0.155 4),the difference was statistically significant(t=3.883 9,P<0.05);SOX2mRNA expression level of the patient’s gender,age,serum AFP level,tumor size was no significant correlation(P>0.05),and TMN staging with clinical correlation.Western Blot shows SOX2 protein in grade Ⅰexpression was significantly lower than the Ⅱ,Ⅲ grade expression.Conclusions: The expression level of SOX2 was up-regulated in HCC tissue as compared with adjacent liver tissue,and is closely related with TMN clinical stage.
