Abstract:Objective To determine the effect of lidocaine on pharmacokinetics of intravenous ketamine in children.Methods Sixteen patients age ranged from 6 to 14 years with ASA Ⅰ underwent ear surgery were randomly received ketamine(2 mg/kg,group A) or combination of lidocaine(1.5 mg/kg) and ketamine(2 mg/kg,group B) during the induction with sevoflurane inhalation.Ketamine plasma levels were determined at 5,15,30,60,120,240 and 480 min after ketamine was administrated using high performance liquid chromatography.Results Ketamine plasma levels were significantly higher at 30,60,120 and 240 min after ketamine administration in group B compared with those in group A.When combined with lidocaine,the mean plasma ketamine concentration-time area under the curve(AUC) value was significantly greater and the clearance was significantly lower when compared with ketamine administrated alone.Moreover,the elimination half-life of ketamine was significantly longer when combined with lidocaine compared with ketamine alone.Conclusion Lidocaine could significantly change the pharmacokinetic of ketamine in children as demonstrated by the elimination half-life prolongation and the clearance reduction.
