Abstract:Objective To observe the expression of soluble resistance-related calcium binding protein(sorcin) in the model of Parkinson's disease(PD) induced by 1-methyl-4-phenyl-pyridinium ion(MPP+) in SH-SY5Y cells,in order to identify proteins related to degeneration of dopaminergic neuron for revealing the potential pathogenesis of PD.Methods The undifferentiated SH-SY5Y cells were treated with 1.0 mmol·L-1 MPP+ for 24 h to set up in vitro PD cell models.Then,quantitative two-dimensional difference in-gel electrophoresis(2D-DIGE) coupled with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS) was performed to identify the differentially expressed proteins in PD models.Results Proteomics analysis revealed that 22 proteins were differentially altered in MPP+-treated SH-SY5Y cells,of which one protein spot was unambiguously identified as sorcin.Sorcin showed a significant increase in level in this PD model,and the fold changes in protein expression between MPP+-treated SH-SY5Y cells and controls was 1.97(P<0.01).Its coverage,expectation and theoretical Mr and pI was 25.7,0.006,21 940 and 5.3,respectively.Conclusion The function role of sorcin is correlated with calcium homeostasis and apoptosis,which may be pertinent in the toxicity induced by MPP+ in SH-SY5Y cells and underlying pathogenesis of PD.
