中药金叶败毒制剂抗人巨细胞病毒感染的作用机制研究

摘    要:目的探讨中药金叶败毒制剂抗人巨细胞病毒(HCMV)感染的实验效果及作用机制.方法用HCMV AD169感染人胚肺成纤维细胞,采用MTT法观察细胞的增殖活性,应用免疫组化方法分别检测中药金叶败毒制剂和更昔洛韦(GCV)干预后感染细胞cyclinD1、细胞周期素依赖激酶4(CDK4)及P16蛋白的表达水平.结果金叶败毒制剂和更昔洛韦能明显促进HCMV感染细胞cyclinD1及CDK 4的表达,降低P16蛋白的表达水平,促进细胞的增殖和分化.结论金叶败毒制剂可能通过提高HCMV感染细胞cyclinD1及CDK 4的表达,降低P16蛋白的表达,促进宿主细胞增殖而发挥抗病毒作用.

Abstract:Objective: To explore the action mechanism of Chinese medicine Jinye Baidu on treating human cytomegalovirus (HCMV) infection. Methods: The cell proliferation activity was observed by MTT and the expressions of the cyclinD1, cell cycle dependent kinase 4(CDK4) and P16 in infected cells were measured by immunohistochemistry before and after treated by Jinye Baidu or Ganciclovir. Results: Both Jinye Baidu and Ganciclovir could increase the expressions of cyclinD1 and CDK4 and decrease the expression of P16 significantly in HCMV infection cells, accelerate the cells proliferation and differentiation. Conclusion: Jinye Baidu agent may play the antivirus action by increasing the expressions of cyclinD1 and CDK4, decreasing the expression of P16 in HCMV infected cells and acceleration host cell proliferation.
