不同保存方法对蚊虫RNA保护效果的评价

目的研究适合口岸媒介生物监测的蚊虫样本保存方法,并对蚊虫RNA保护效果进行评价。方法采用普通冰箱、低温冰箱、液氮、RNALater等保存方式,对蚊虫进行1周、1个月、6个月保存,提取蚊虫RNA,比较这些保存方式对蚊虫RNA保护效果。结果 RNALater与液氮对蚊样本均具有良好的保存效果,在1周、1个月、6个月内,蚊虫RNA均未见明显降解;低温冰箱在1周、1个月内对蚊样本有较好的保存效果,蚊虫RNA未见明显降解,但6个月后,蚊虫RNA降解明显;常温冰箱(-20℃)保存效果不佳,仅在1周之内对蚊虫核酸有一定的保护效果,1个月后,RNA发生明显降解。结论 RNALater、液氮和低温冰箱在1个月内对蚊虫RNA均具有较好的保存效果,但RNALater具有保存时效长、使用方便等优点,更适用口岸蚊虫监测中样本的保存和运输。

The evaluation of the protective effect of different preservation methods to mosquito RNA
