人骨肉瘤中5个新基因片段在不同骨肿瘤中的差异表达

摘    要:目的 筛选数个在人骨肉瘤中检出率高的差异表达基因片段并探讨其临床意义。方法 以筛选出5个新基因片段作为研究对象,采用RT-PCR技术检测37例骨肉瘤,68例其他骨肿瘤及19例截除肢体上的正常骨组织标本。结果 在37例骨肉瘤中,表达检出率高的5个片段依次为:ZOL03为92.6%,1C82为89.4%,1C74为73.9%,2G12为66.5%,2A21为53.7%;在23例骨软骨瘤中表达率依次为:ZOL03为6.5%,1C82为14.7%,1C74为8.3%,2G12为11.9%,2A21为12.2%;在29例骨巨细胞瘤中表达率依次为:ZOL为8.3%,1C82为15.6%,1C74为12.5%,2G12为13.8%,2A21为5.4%;在16例软骨肉瘤中表达率依次为:ZOL03为9.4%,1C82为14.3%,1C74为7.5%,2G12为6.8%,2A21为6.2%;在19例正常骨组织标本中表达率依次为:ZOL03为0,1C82为6.4%,1C74为0,2G12为4.8%,2A21为3.9%,这5个基因片段检出率在骨肉瘤与其他骨肿瘤中存在显著差异(P<0.01);其检出率与骨肉瘤与正常骨组织中存在显著差异(P<0.01)。结论 ZOL03,1C82,1C74,2G12及2A21这5个基因片段可能与骨肉瘤的发生,发展有关,监测骨组织中此5个基因片段的表达水平的变化可能对骨肉瘤的早期发现更有意义。

Abstract:AIM To screen several genes differentially expressed in human osteosarcoma with higher examination rates and discuss its clinical significance. METHODS By using ZOL03, 1C82, 1C74, 2G12 and 2A21 as research targets, 37 specimens of human osteosarcoma and 68 specimens of other musculoskeletal tumors as well as 19 specimens were screened by quantitative RT PCR. RESULTS In 37 specimens of osteosarcoma, percentages of the 5 fragments with higher examination rate were 92.6% (ZOL03), 89.4% (1C82), 73.9% (1C74), 66.5% (2G12) and 53.7% (2A21) sequentially. In 23 specimens of osteochondroma, percentages of the 5 fragments with higher examination rate were 6.5% (ZOL03), 14.7% (1C82), 8.3% (1C74), 11.9% (2G12) and 12.2% (2A21) respectively. In 29 specimens of giant cell tumor of bone, the percentages of the 5 fragments in turn were 8.3% (ZOL03), 15.6% (1C82), 12.5% (1C74), 13.8% (2G12) and 5.4% (2A21). In 16 specimens of chondrosarcoma, the percentages in succession were 9.4% (ZOL03), 14.3% (1C82), 7.5% (1C74), 6.8% (2G12) and 6.2% (2A21). The expression rates of the 5 fragments between osteosarcoma and other musculoskeletal tumors were significantly different ( P <0.01). And in 19 specimens of normal bone tissue, the expression rates of the 5 fragments were 0 (ZOL03), 6.4% (1C82), 0 (1C74), 4.8% (2G12) and 3.9% (2A21). The expression rates of the 5 fragments between osteosarcoma and normal bone tissue were significantly different ( P <0.01). CONCLUSION The 5 fragments of differentially expressed genes isolated from human osteosarcoma tissue may be correlated with occurrence and progress of osteosarcoma.
