外源NGF促进大鼠皮肤创面模型损伤修复的机制研究

摘    要:目的:探究外源NGF对大鼠创面的治疗作用及机制。方法:在SD雄性大鼠的背部脊柱的两侧手术切开形成两个2 cm × 2 cm的伤口,右边伤口处给予20 mg/ml NGF 1 ml,左边伤口给等量生理盐水作为对照组。隔天给药到第14天(第14天停止给药),分别于d3、d7、d14统计创面伤口愈合率,并取这3个时间点的组织用免疫组化的方法检测转化生长因子( transforming growth factor-β1,TGF-β1)、CD68-阳性巨噬细胞、增殖性细胞核抗原(proliferating cell nuclear antigen,PCNA)、血管内皮生长因子(vascular endothelial growth factor,VEGF-1)的表达量。结果:NGF组的创面愈合率明显高于生理盐水组,并且NGF组TGF-β1、CD68、PCNA、VEGF-1的蛋白表达量也显著高于生理盐水组(P〈0.05)。结论:外源NGF具有促进创面愈合作用,其机制可能与趋化炎症因子、加快血管生成、促进PCNA、TGF-β1的表达相关。

Abstract:Objective: To study the therapeutic effect of nerve growth factor (NGF) on wound healing in rats and its mechanism. Methods:Two full thickness wounds (2 cm ×2 cm) extending through the panniculus carnosus were made on both sides of the back spine in SD male rats. The right incision (designated as an experimental group) was treated with 1 ml of NGF (20 mg/ml, dissolved in 0.9%sterile saline) every other day till 14th days while the left incision (designated as a control group) with equal amounts of 0.9%sterile saline. The rates of wound healing were recorded at day 3, 7 and 14 and the expression of transforming growth factor-β1 (TGF-β1), CD68+macrophages, proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF-1) in wound tissues sampled at day 3,7 and 14 were detected by immunohistochemistry staining. Results:The rates of wound healing and the protein expression of TGF-β1、CD68、PCNA、VEGF-1 were obviously higher in the experimental group than in the control group (P〈0.05). Conclusion:The mechanism of NGF in the promotion of wound healing might be related to the chemotactic factor, the promotion of angiogenesis and the expression of PCNA and TGF-β1.
