摘    要:目的:建立藁本内酯的RP-HPLC分析方法,并应用于藁本内酯在小鼠体内各组织的分布研究。方法:生物样品经过正己烷萃取浓集后,以尼莫地平为内标,用Kromasil C18柱(150mm×4.6mm,5μm)进行分析,以乙腈-10%异丙醇水溶液(58:42)为流动相,流速1.0mL/min,在324nm处进行检测。结果:藁本内酯与内标的保留时间分别为7.2min和5.9min,藁本内酯在各组织中一定浓度范围内线性关系良好,各组织中低、中、高浓度蒿本内酯的回收率及精密度均符合方法学要求。藁本内酯在小鼠主要效应器官中浓度分布如下:c肺〉c心〉C脑,在主要消除器官中c肝〉c脾〉c肾。藁本内酯在肺和脾脏中分布较多。结论:本方法方便、准确、可靠、可用于藁本内酯的体内分析研究。

Abstract:Objective: To establish a HPLC method for the determination of ligustilide, and utilize it for the tissue distribution study in mice. Methods: After extracted with n-hexane, the sample was added with nimodipine (internal standard). The Kromasil C18column(150 mm × 4.6 μm, 5 μm) was used with the mobile phase consisting of acetonitrile and 10% isopropanol (58 : 42). The flow rate was 1.0 mL/min and the detection wavelength was 324 nm. Results:The retention time of ligustilide and nimodipine was 7.2 min and 5.9 min, respectively. The linear correlation of ligustilide in different tissues was fine. The recovery and precision were in line with the requirements of methodology. The distributions of ligustilide in main effector organs were as follows: c lung〉c heart〉c brain, and distrabutions in main eliminator organs were as follows: c liver〉c spleen 〉c kidney. The concentration of ligustilide was much higher in mouse lung and spleen than other organs. Conclusion: This HPLC method for determination of ligustilide in bio-samples is convenient, accurate and reliable and could applied in the analysis of ligustilide in vivo.
