G蛋白β_3亚单位基因C825T多态性影响缬沙坦的降压疗效

摘    要:目的探讨G蛋白β3亚单位基因C825T多态性与缬沙坦的降压疗效的关系。方法采用聚合酶链反应限制片段长度多态性方法检测147例健康人(对照组)和321例高血压病患者(高血压组)的G蛋白β3亚单位C825T多态性,其中102例高血压病患者口服缬沙坦4周。结果高血压组G蛋白β3亚单位C825T多态性中基因型频率(CC28.7%、CT 52.0%、TT 19.3%)、等位基因频率(C 54.7%、T 45.3%)与对照组基因型频率(CC 27.2%、CT 46.9%、TT25.9%)、等位基因频率(C 50.7%、T 49.3%)比较无显著性差异;缬沙坦对CT(18.29±11.17)mm Hg,1 mm Hg=0.133 kPa]、TT(25.63±22.68)mm Hg]、CT+TT(19.25±13.20)mm Hg]基因型患者降低收缩压的作用强于CC基因型(11.33±9.15)mm Hg,P<0.05];对CT(15.03±9.35)mm Hg]、CT+TT(14.50±9.23)mm Hg]基因型患者降低舒张压的作用强于CC基因型(8.81±5.60)mm Hg,P<0.05]。结论G蛋白β3亚单位基因C825T多态性与缬沙坦的降压疗效相关,而与原发性高血压无关。

Abstract:Objective To investigate the relationship between G protein β_3 subunit gene polymorphism and the response to antihypertensive treatment with valsartan.Methods Polymerase chain reaction combined with restriction enzyme digestion was used to detect the polymorphism of G protein β_3 subunit gene(GNB3 gene) in 147 normotensive controls and 321 hypertensive patients.102 hypertensive patients were treated with valsartan for 4 weeks.Results There was no significant difference in the GNB3 gene C825T polymorphism genotypes and alleles between hypertensive patients(CC 28.7%,CT 52.0%,TT 19.3%,C 50.7%,T 49.3%) and normotensive controls(CC 27.2%,CT 46.9%,TT 25.9%,C 50.7%,T 49.3%).Mean decreases in systolic blood pressure were significantly greater in CT,TT,CT+TT genotypesCT(18.29±11.17) mm Hg,TT((25.63)±22.68) mm Hg,CT+TT(19.25±13.20) mm Hg]than in CC genotype (11.33±9.15) mm Hg,P<0.05].Mean decreases in diastolic blood pressure were significantly greater in CT (15.03±9.35)(mm Hg)] and CT+TT(14.50±9.23) mm Hg] genotypes than in CC genotype(8.81±5.60) mm Hg,(P<)0.05].Conclusion The C825T polymorphism of G protein β_3 subunit is related to the response to valsartan but not to essential hypertension.
