利用应激建立抑郁相关动物模型

目的 :依据应激理论结合现代实验动物学 ,建立病、“证”相结合的动物模型 ,探讨抑郁的基本病机。方法:利用环境刺激造成应激状态后加半量利血平造成抑郁动物模型 ,采用高效液相色谱 -电化学技术检测各组小鼠全脑去甲肾上腺素 (NE)、五羟色胺 (5 -HT)的含量,并与经典的抑郁模型组、半量利血平组、空白组比较。结果:与空白对照组、半量利血平组相比 ,模型组小鼠脑内NE、5 -HT含量显著下降 ,具有统计学意义 (P<0.01) ;与经典抑郁模型组相比无显著性差异 (P>0.05)。结论:应激加半量利血平所造动物模型成功 ,说明环境刺激引起的应激所致气机失调与抑郁的发生、中枢NE、5 -HT等神经递质变化、神经内分泌调节轴有密切的相关性

Establishment of animal models for depression by stress

Objective: To research the mechanism of depression.Methods:Models are made by full amount of reserpine injection,half amount of reserpine and half amount of reserpine combined with stimulating which is called Qi dynamic model.The concentration of NE and 5-HT is measured by the methods of HPLC electro-chemistry.Results:Compared to the normal group and half amount of reserpine group, the concentration of NE and 5-HT in the rat brain of two models is obviously decreased (P<0.01),while there is no obvious difference between the two models themselves. Conclusion: Qi dynamic model is made successfully.Qi dynamic disorders syndrome has close relation with depression and concentration changes of NE and 5-HT.
