| 摘 要： | 目的:研究受体胸腺内注射Fas抗体对心脏移植大鼠Fas/FasL表达的影响。方法:应用大鼠同种异位腹腔心脏移植模型,将模型动物随机分为三组,每组8只动物。A组:移植前后不给予处置;B组:给环孢素A,按每日8mg/kg于术前1d口服;C组:胸腺注射Fas抗体,以0.1mg/kg剂量在术前注射。以免疫组化法和逆转聚合酶链式反应(RT-PCR)法观察术后移植心脏局部的Fas及FasL的表达情况。结果:与A组相比,受体胸腺内注射Fas抗体可减少移植心脏局部的Fas/FasL基因表达(RT-PCR法半定量结果P0.05)和蛋白表达(免疫组化法P0.05)。结论:受体胸腺内注射Fas抗体是一种有效的抑制心脏移植急性排斥反应的方法,可使移植心肌局部Fas/FasL表达下调。 |
