椒目油对哮喘大鼠血清总IgE、IL-13水平及肺组织Fas/FasL表达的影响

摘    要:目的观察椒目油(zanthoxylum seed oil,ZSO)对支气管哮喘大鼠模型血清中总IgE、IL-13水平及肺组织Fas/FasL表达的影响,为临床应用椒目油治疗哮喘提供实验证据和理论支持。方法将雄性SD大鼠60只随机分为正常对照组、哮喘模型组、地塞米松治疗组和椒目油治疗组,每组各15只。以卵白蛋白(ovalbumin,OVA)腹腔注射致敏和雾化吸入激发建立哮喘大鼠模型。收集支气管肺泡灌洗液(BALF),行细胞计数;采用ELISA法检测血清中总IgE和IL-13的含量;以免疫组化ABC法检测肺组织中Fas/FasL蛋白的表达。结果椒目油和地塞米松均能降低哮喘肺组织BALF中白细胞、嗜酸性粒细胞和淋巴细胞的数量,与哮喘模型组相比较,有统计学意义(P〈0.01)。与哮喘组相比,椒目油和地塞米松均能显著下调哮喘大鼠血清中总IgE和IL-13的含量(P〈0.05),并增强肺组织淋巴细胞表面Fas/FasL的蛋白表达(P〈0.05)。结论椒目油可抑制哮喘大鼠的免疫细胞产生IgE和IL-13,减轻Eos浸润,并可能通过增强Fas/FasL蛋白表达而促进炎症局部淋巴细胞凋亡,具有明显的抗炎、抗过敏作用。

Abstract:Objective To study the effect of zanthoxylum seed oil (ZSO) on serum total IgE, interleukin 13 (IL- l3) and Fas/FasL in lung tissue of asthmatic rat. Methods The rats were divided into four groups, including normal control group (A), asthmatic model group (B), dexamethasone treatment group (C) and zanthoxylum seed oil treatment group (D). The model of asthmatic rat was constructed with immunization of intraperitoneal injection with saline ovalbumin (OVA) and challenge of inspiration with nebulized OVA. The level of serum IL-13 and total IgE were detected with ELISA. Protein expressions of Fas/FasL in lung tissues were assayed by immunohistochemistry. The number of leuco-cyte (WBC) in bronehoalveolar lavage fluid (BALF) were counted. Results The number of WBC, eosinophile granulo- cyte and lymphocyte in BALF in group C and D were decreased significantly (P〈0.01) compared with group A. The concentration of serum IL-13 and total IgE of group C and D were significantly lower than those of group B (P〈0.05). The protein expressions of Fas/FasL in lung tissue in group C and D were significantly higher than that in group B(P〈 0. 05). Conclusions Zanthoxylum seed oil can decrease the level of serum total IgE, IL-13 and promote apoptosis of local lymphocyte by increasing gene expression of Fas/FasL to antagonize asthmatic airway inflammation.
