| 摘 要： | 目的:建立一种耐药性稳定的人涎腺黏液表皮样癌裸鼠移植瘤模型.方法:将人涎腺黏液表皮样癌多药耐药细胞接种于裸鼠皮下,成瘤后经腹腔注射小剂量5-FU进行体内诱导,建立稳定的黏液表皮样癌裸鼠移植瘤模型.结果:裸鼠移植瘤原代培养的MEC/5-FU/NU细胞与体外培养细胞结构类似,MEC/5-FU/NU细胞对5-FU的耐药指数为27.82.RT-PCR显示耐药相关基因ABCB1、ABCB11、GSTA1在MEC/5-FU/NU细胞中明显高表达(P<0.05).免疫荧光显示多药耐药蛋白MDR-1在MEC/5-FU/NU细胞中高表达.结论:以MEC/5-FU建立的黏液表皮样癌裸鼠皮下移植瘤模型仍保持MEC/5-FU细胞的形态学特征和多药耐药性,可为耐药性人涎腺黏液表皮样癌耐药机制和逆转的研究提供较理想的动物模型. |
