| 摘 要： | 目的观察顺铂作用肺癌A549细胞后自噬的发生,及肺岩宁对自噬相关蛋白Atg5、Atg7的影响。方法顺铂和肺岩宁处理肺癌A549细胞,CCK-8筛选顺铂和肺岩宁的优化给药浓度,CCK-8检测各组A549细胞增殖能力,Western blot检测各组自噬蛋白表达。结果与单药顺铂相比,3 mg/L的顺铂联合20 mmol/L的自噬抑制剂氯喹明显提高了A549细胞增殖的抑制率(P0.05)。同时,顺铂导致了微管相关蛋白1轻链3Ⅱ型(microtubule-associated protein 1 light chain 3,LC3Ⅱ)蛋白表达增加(P0.01)。肺岩宁干预A549细胞后,自噬蛋白Atg5蛋白表达低于模型组和顺铂组(P0.05),Atg7蛋白表达显著低于模型组和顺铂组(P0.01)。结论顺铂诱导了A549细胞产生保护性自噬,肺岩宁可能通过抑制肺癌细胞自噬来发挥抗肺癌细胞增殖作用。
