骨痛灵方调控骨转移模型小鼠脊髓MAPK信号通路的研究

摘    要:目的从调控MAPK信号通路角度探讨骨痛灵方治疗骨转移疼痛的作用机制。方法将40只小鼠随机分为假手术组、模型组、中药组、对照组,除假手术组外,其余各组建立小鼠骨转移模型。从造模第7天开始给药,中药组予骨痛灵方治疗,对照组以曲马多治疗,采用VonFrey纤维检测机械痛阈值评价其止痛疗效,观察肝肾心病理评价其安全性,采用Western blot法检测脊髓MAPK信号通路中p-JNK、p-p38、p-ERK蛋白的表达。结果 (1)造模第14天,与模型组比较,对照组和中药组机械痛阈值均有升高(P0.05)。造模第21天,中药组机械痛阈值较模型组与对照组均增高(P0.05)。(2)各组心肝肾病理均无明显改变。(3)与假手术组比较,模型组脊髓p-JNK、p-p38、p-ERK的表达均增加(P0.05)。与模型组比较,对照组和中药组脊髓p-JNK、p-p38和p-ERK的表达均显著减少(P0.05)。中药组和对照组脊髓p-JNK、p-p38和p-ERK的表达差异均无统计学意义(P0.05)。结论骨痛灵具有良好的治疗骨转移疼痛的疗效,且对心肝肾无明显毒副作用,其止痛作用机制与调控MAPK信号通路p-JNK、p-p38、p-ERK蛋白表达有关。
