摘    要:目的:探讨PTEN基因表达水平及其基因突变与脑胶质瘤的发生及恶性进展的关联性。方法:选取脑胶质瘤组织15例,以4例非胶质瘤脑组织为对照,采用逆转录聚合酶链反应(RT-PCR)法检测脑胶质瘤组织PTEN mRNA表达水平,采用单链构象多态性分析法(SSCP analysis)快速筛查脑胶质瘤组织PTEN基因突变状况。结果:RT-PCR结果,脑胶质瘤组织PTEN mRNA表达水平(0.063±0.006)较对照组(0.126±0.015)明显降低(P<0.05)。经单链构象多态性分析,脑胶质瘤组织PTEN基因外显子8、9未检测到突变,但其中1例PTEN基因外显子5的聚丙烯酰胺凝胶电泳结果显示电泳带飘移,考虑可能为杂合子突变。结论:PTEN基因低表达及基因突变可能对胶质瘤发生、发展有促进作用。

Abstract:Objective To study the relationships between the expression level of PTEN gene, gene mutation and occurrence and development of glioma.Methods Fifteen glioma tissues were selected,and the normal 4 non-glioma tissues were used as control.The expression level of PTEN mRNA was measured by RT-PCR method,and the mutation of PTEN gene was screened by SSCP analysis in various histopathological grades of glioma.Results Compared with control group(0.126±0.015),the level of PTEN mRNA in glioma tissues(0.063±0.006) was decreased(P<0.05).SSCP analysis showed that there was not mutation of the bands of exon 8 and 9 of PTEN gene .The band shift of exon 5 of PTEN gene of one specimen was found on ployacrylamide gel electrophoresis.Conclusion The low expression and mutation of PTEN gene probably promote the occurrence and development of glioma.
