肠缺血再灌注损伤后肠上皮RSpo1表达及其意义

目的 探讨R-Spondinl(RSpol)在肠缺血再灌注损伤后肠道的表达及其意义.方法 健康雄性昆明小鼠50只,分为对照组(10只)和实验组(40只).对照组小鼠仪作剖腹手术,实验组小鼠麻醉后夹闭肠系膜动脉20min后松夹,分为再灌流6 h(A组)、12 h(B组)、24 h(C组)和48 h(D组).采用ELISA和RT-PCR方法检测小肠组织RSpol的表达.结果 RT-PCR和ELISA结果均显示RSpol在肠缺血再灌注损伤后6h表达较对照组显著降低(P<0.05),12 h表达逐渐增加,在24 h达到最高并显著高于对照组(P<0.05),随后其表达迅速下降,在48 h显著低于对照组(RT-PCR:P<0.05:ELISA:P<0.01).结论 缺血再灌注损伤能抑制肠道内分泌细胞早期RSpol表达,诱导中期RSpol表达,RSpol能促进肠道上皮干细胞增殖分化,参与肠黏膜受损后的修复过程.
