奥曲肽联合奥美拉唑体外对胃癌细胞株SGC-7901生长的影响

目的探讨联合奥曲肽与奥美拉唑在体外对人胃癌细胞株SGC-7901增殖的影响及其可能的作用机制。方法采用MTT法检测奥曲肽和奥美拉唑对SGC-7901细胞生长的影响,根据中效原理判断联合用药的效果;流式细胞仪检测联合用药对SGC-7901细胞的细胞周期、Bcl-2和Bax蛋白表达的影响。结果奥曲肽联合奥美拉唑在体外能显著抑制SGC-7901细胞的生长,联合用药对SGC-7901细胞的抑制作用强于单独用药,两药联合应用在高水平效应合用指数小于1,具有明显的协同作用;联合用药能使SGC-7901细胞周期停滞于G0/G1期,下调Bcl-2且上调Bax蛋白的表达。结论奥曲肽联合奥美拉唑在体外可以抑制SGC-7901细胞的生长,具有协同效应,其机制可能与下调Bcl-2蛋白和上调Bax蛋白表达有关。

Effects of octreotide combined with omeprazole on proliferation of SGC-7901 cells in vitro

Objective To investigate the effect and mechanism of octreotide combined with omeprazole on proliferation of gastric cancer cell line SGC-7901.Methods The effect on proliferation in SGC-7901 was determined by MTT methond,and the combination effect of two agents was evaluated according to the median-response principle.The cell cycle and the expression of Bcl-2 and Bax protein of SGC-7901 were all detected by flow cytometry.Results Both octreotide and omeprazole could respectively inhibit the proliferation of SGC-7901 cell,and the effect of combination use was better than that of single use,the combination indexes for the two drugs were less than 1 in high responses.Octreotide combined with omeprazole could induced the cells arrest at G0/G1 stage,up-regulate the expression of Bcl-2 and down-regulate the expression of Bax.Conclusion Octreotide combined with omeprazole could increase the inhibition effect significantly on proliferation of SGC-7901 cell in vitro,and the mechanism might be associated with the regulation on expression of Bcl-2 and Bax proteins.
