AT2受体诱导新生大鼠肾小管上皮细胞凋亡的研究

目的 探讨AT_2受体在诱导肾小管上皮细胞凋亡中的作用。方法 通过取新生大鼠肾小管上皮细胞体外培养,传代后加入不同浓度AT_2受体兴奋剂CGP-42112A,在不同时间用流式细胞仪测定凋亡细胞比值,并结合TUNEL分析。结果CGP-42112A不同浓度加入细胞24h后,随着剂量的增大,凋亡细胞所占比值逐渐增加,与对照组比较均具有显著差异,P<0.001。10~(-5)M CGP-42112A作用于细胞,随着时间的延长,凋亡细胞所占比值逐渐增加,与对照组比较有显著差异,P<0.001。结论 AT_2受体可诱导新生大鼠肾小管上皮细胞凋亡,凋亡细胞数与受体剂量正相关,与受体兴奋剂作用时间正相关。

STUDY OF ANGIOTENSIN II AT_2 RECEPTOR INDUCED RENAL TUBULAR CELLS APOPTOSK

Objective To investigate the role of Angiotensin II AT2 receptor in renal tubular cells apoptosis. Methods In vitro transfered and cultured renal tubular cells AT2 receptor agonist CGP - 42112A in different doses were diffused, then cell apoptosis was analyzed by TUNEL and flow cytometry. Results With the diffusion of CGP - 42112A in different doses per group respectively (n = 4) , after 24 h, apop-totic cells rations shown difference with the controls, P < 0.001. For CGP - 42112A 10-5 M diffused cells, after 24h, 48h, 60h, 72 h, apoptotic cells were increased compared with control group, P < 0.001. Conclusion AT2 receptor may induce the apoptosis of renal tubular cells. CGP - 42112A has positive dose and time dependent effect on cells apoptosis.
