维药异常黑胆质成熟剂对大鼠乳腺增生模型的影响

摘    要:目的:观察维药异常黑胆质成熟剂对实验性乳腺增生模型大鼠乳腺组织形态学变化、脏器指数、血清性激素水平的影响。方法选用雌性 Wistar 大鼠60只,随机分为正常对照组、模型对照组、阳性对照组(三苯氧胺组,1.8 mg/kg)及异常黑胆质成熟剂高、中、低剂量组(15.00、7.50、3.75 g/kg)。采用大鼠右下肢肌肉注射雌、孕激素建立乳腺增生病模型,分别用异常黑胆质成熟剂高、中、低剂量及三苯氧胺片连续给药45 d。常规 HE 染色,光镜下观察乳腺组织病理形态;计算脾脏、胸腺、子宫及卵巢指数;采用放射免疫法检测其血清雌二醇(E2)、孕酮(P)、睾酮(T)水平;用 GPO-PAP 法测定甘油三酯(TG)含量;用 PTA-Mg2+沉淀法测定高密度脂蛋白(HDL-C)含量;用 PVS 沉淀法测定低密度脂蛋白(LDL-C)含量;用 CHOD-PAP 法测定总胆固醇(TC)含量。结果与模型对照组比较,阳性对照组及异常黑胆质成熟剂中、高剂量组乳腺导管管腔变小,导管上皮细胞增生程度减轻,结缔组织增生程度亦减轻,异常黑胆质成熟剂高剂量组部分可接近正常大鼠乳腺结构。与模型对照组比较,异常黑胆质低、中剂量组脾脏、胸腺指数(P <0.05)降低,异常黑胆质低、中、高剂量组子宫及卵巢指数降低(P <0.05);与模型对照组比较,异常黑胆质低、中、高剂量组血清 E2降低、P、T 水平升高(P <0.05~0.01),而阳性对照组与模型对照组及空白对照组比较,P、T 水平未见升高;与模型对照组比较,异常黑胆质成熟剂高、中、低剂量组血清 TC、TG、LDL-C、HDL-C 水平均不同程度地降低(P <0.05~0.01)。结论异常黑胆质成熟剂可改善增生乳腺的病理形态变化,调节模型大鼠血清性激素和血脂代谢紊乱状态,能够对抗雌激素引起的大鼠乳腺增生。

Abstract:Objective To observe the effects of Uyghur Medicine Abnormal Savda Munziq (ASMq)on pathologic changes of breast tissue,visceral indexes,serum sex hormone levels in experimental rat model of mammary gland hyperplasia.Methods 60 rats were randomly divided into 6 groups that,blank control group,model control group,positive control (tamoxifen)group,ASMq high-dose group,ASMq middle-dose group,ASMq low-dose group.The rat model of mammary gland hyperplasia were established by tak-ing estradiol benzoate and progesterone,The treatment groups were given with different doses of ASMq and tamoxifen pills for 45 days (ASMq high,middle and low dose respectively 15.00,7.50,3.75 g/kg, tamoxifen 1.8 mg/kg).Routine HE stained and pathological morphology of breast tissue were observed through optical microscope;Visceral index(thymus,spleen,uterus and ovary)were determined;the con-centration of estradiol (E2 ),progesteron(P),testosterone(T)were detected by radioimmunoassay;The content of triglyceride (TG),high density lipoprotein(HDL-C),low density lipoprotein cholesterol (LDL-C),total cholesterol (TC)were respectively determined by CHOD-PAP method,PTA-Mg2+ precipitation method,PVS precipitation method and GPO-PAP method.Results Compared with model control Groop, in positive control group and ASMq middle-dose and high-dose group,mammary gland catheter were smal-ler,the degree of ductal epithelial hyperplasia and fibrous tissue hyperplasia were reduced,part of high-dose group mammary gland structure were close to normal;Compared with model control Group,thymus and spleen index were decreased in ASMq low and middle-dose group (P 〈0.05),uterus and ovary index were decreased (P 〈 0.05 )in ASMq high,middle and low-dose group;Compared with model control group,ASMq decreased the concentration of E2 ,elevated the level of P and T (P 〈 0.05 ~ 0.01),al-though,compared with model control group and blank contral group,there were no recovery of P and T level in positive control group
