中国人G6PD基因EcoRI酶切位点研究

摘    要:应用DNA Southern印迹杂交技术,以G6PD cDNA为探针,研究了16种限制酶的限制性片段长度多态,结果表明除EcoRⅠ外,其余15种限制酶均无多态改变,分析39例正常人与38例G6PD缺乏者G6PD基因ECO RⅠ酶切位点时,发现约20%的人出现7·0 kb的片段,1例G6PD患者出现6.0 kb片段,国外并无类似报道,并对这些片段来源及意义进行初步探讨。

Abstract:The restriction fragment length polymorphisms (RFLP) of sixteen restriction endonuc-leases in G6PD gene were identified by using Southern blotting. Fifteen endonucleases were found no changes except for EcoR I . The results of 39 cases of normal individuals and 38 cases of G6PD deficients for G6PD gene digested by EcoR I showed that 20% individuals revealed 7.0 kb band, one G6PD deficient was 6.0 kb band, which were different from the abroad reports. The origin and significance for these fragments were discussed in this paper.
