乌梅丸拆方对溃疡性结肠炎大鼠细胞因子的影响及其寒热配伍的意义研究

摘    要:目的从乌梅丸拆方对溃疡性结肠炎模型大鼠细胞因子含量变化的影响来探讨该方寒热配伍作用机制及配伍意义。方法将SD雄性大鼠分为空白对照组、模型组、乌梅丸组、寒热并用组、温热组、寒凉组,除空白对照组外其余各组采用苦寒药物灌胃加TNBS乙醇灌肠的方法,诱导寒热错杂型溃疡性结肠炎动物模型,各组给予相应的干预措施。检测大鼠结肠组织IL-1、IL-10、TNF-α、PGE2、MPO含量变化及血清白细胞计数;HE染色行病理组织学观察。结果与空白对照组比较,模型组大鼠结肠组织TNF-α、IL-1、MPO、PGE2、血清白细胞计数含量显著升高,IL-10水平显著降低,差异有统计学意义(P<0.05)。与模型组比较,乌梅丸组、寒热并用组、温热组TNF-α、IL-1、MPO、PGE2、血清白细胞计数均降低,IL-10水平升高,而寒凉组TNF-α、IL-1、MPO、血清白细胞计数均降低,差异有统计学意义(P<0.05)。与乌梅丸组比较,寒热并用组、温热组和寒凉组TNF-α、IL-1较高,而IL-10较低,温热组和寒凉组MPO较高,寒凉组PGE2较高,温热组白细胞计数较高,差异均有统计学意义(P<0.05)。与寒热并用组比较,温热组和寒凉组TNF-α、IL-1较高,温热组MPO、IL-10、白细胞计数较高,寒凉组IL-10较低、PGE2较高,差异有统计学意义(P<0.05)。与温热组比较,寒凉组TNF-α、IL-1、MPO、IL-10、白细胞计数降低,PGE2含量升高,差异有统计学意义(P<0.05)。结论温热组药物在升高IL-10和降低PGE2水平方面作用突出;寒凉组药物在降低IL-1、TNF-α、MPO含量和血清白细胞计数,对抗体温升高方面作用突出;乌梅丸通过寒热配伍既能消除单用寒凉药或温热药的不利作用,又有协同和增效作用。

Abstract:Objective To discuss the significance and mechanism of herbal compatibility from the effect of decomposed "Wumei Pill" on cytokines in rats with ulcerative colitis.Methods The male SD rats were randomly divided into blank group,model group,"Wumei Pill" group,combined cold-heat group,heat group and cold group.Except for the blank group,rats in the other groups were given cold medicine by intragastric administration and TNBS ethanol by enema to establish ulcerative colitis models,and each group was intervened by corresponding treatments.The contents of IL-1,IL-10,TNF-α,PGE2 and MPO in colon tissues of rats were detected and the WBC was measured;observe the indicators pathologically by HE staining.Results Compared with the blank group,the contents of TNF-α,IL-1,MPO,PGE2 and WBC were increased in model group,and the IL-10 level was decreased,with significant differences between blank group and model group(P<0.05).Compared with the model group,TNF-α,IL-1,MPO,PGE2 and WBC in "Wumei Pill" group,combined cold-heat group and heat group were decreased,and the IL-10 level was increased;while in cold group,the TNF-α,IL-1,MPO and WBC were decreased,with significant differences between model group and the other groups(P<0.05).Compared with "Wumei Pill" group,the TNF-α and IL-1 were higher in combined cold-heat group,heat group and cold group,but the IL-10 was lower.MPO in heat group and cold group,PGE2 in cold group and WBC in heat group were all higher,with significant difference between "Wumei Pill" group and the other groups(P<0.05).Compared with combined cold-heat group,the TNF-α and IL-1 in heat group and cold group,MPO,IL-10 and WBC in heat group and IL-10,PGE2 in cold group were all higher,with significant differences between combined cold-heat group and the other groups(P<0.05).Compared with heat group,TNF-α,IL-1,MPO,IL-10 and WBC in cold group were decreased,and PGE2 was increased,with significant differences between them(P<0.05).Conclusion Medicines in heat group can elevate IL-10 and lower PGE2;medicines in cold group can decrease IL-1,TNF-α,MPO,WBC and can lower the temperature.Compatibility of cold and heat herbs can eliminate their adverse effects and enhance the cooperative effect.
