升降散对内毒素刺激血管内皮细胞ET-1和NO表达的影响

摘    要:目的:探讨升降散对内毒素(LPS)刺激下血管内皮细胞ET-1和NO表达的影响。方法:将内皮细胞传代培养后分为空白组、对照组(空白细胞+LPS 0.50 mg/ml)、低剂量升降散组(升降散5 mg/ml+LPS 0.50 mg/ml)、中剂量升降散组(升降散10 mg/ml+LPS 0.50 mg/ml)和高剂量升降散组(升降散30 mg/ml+LPS 0.50 mg/ml),观察各组ET-1和NO水平变化。结果:升降散作用细胞48 h后,内皮细胞在5~30 mg/ml范围内对内皮细胞贴壁无明显影响。LPS作用内皮细胞48 h后,NOi、NOS、NOS水平明显升高,与空白组比较有显著性差异(P<0.01);升降散作用细胞后ET-1、NOi、NOS、NOS水平有不同程度降低,与对照组比较有显著性差异(P<0.05,P<0.01),且中、高剂量升降散组诸指标的降低幅度较低剂量升降散组明显(P<0.05,P<0.01)。结论:升降散能抑制LPS诱导的VECs活性物质生成和释放,从而减轻对组织细胞的损伤,以中高剂量升降散效果更显著。

Abstract:Objective: To study the effects of "Shengjiang Powder" on lipopolysaccharide(LPS)-stimulated ET-1 and NO in vascular endothelial cells(VECs).Methods: The cultured endothelial cells were divided into five groups: normal group,control group(LPS 0.50 mg/ml),low-dose "Shengjiang Powder" group("Shengjiang Powder" 5mg/ml+LPS 0.50 mg/ml),middle-dose Shengjiang Powder group("Shengjiang Powder" 10 mg/ml +LPS 0.50 mg/ml)and high-dose "Shengjiang Powder" group("Shengjiang Powder" 30 mg/ml+LPS 0.50 mg/ml).The levels of endothelin-1(ET-1) and nitric oxide(NO) were detected and compared in all groups.Results: Within the concentrations of 5-30 mg/ml,"Shengjiang Powder" did not act on the VECs.After LPS acting on VECs for 48 hours,the levels of ET-1,NO,iNOS and NOS in cell culture medium were significantly increased,with striking differences compared with control group(P<0.01).In the three "Shengjiang Powder" groups,the levels of ET-1,NO,iNOS and NOS were significantly decreased compared with LPS group(P<0.05,P<0.01);and the decrease of ET-1,NO,iNOS and NOS was more significant in the high-and middle-dose "Shengjiang Powder" group than in low-dose "Shengjiang Powder" group(P<0.05,P<0.01).Conclusion: "Shengjiang Powder" can inhibit the production and release of LPS-stimulated VECs to relieve tissue injury,with better results in higher concentration of Shengjiang Powder.
