大黄及大黄素对大鼠胰腺细胞外基质降解作用的影响

摘    要:目的从胰腺细胞外基质降解环节探讨大黄及大黄素对模型大鼠血清MMP-2、MMP-9及TIMP-1含量的影响。方法将SD大鼠随机分为假手术组、模型组、大黄治疗组、大黄素治疗组4组,每组10只。结扎胰胆管并腹腔注射雨蛙素50 ng/g建立大鼠胰腺细胞外基质沉积动物模型,造模前后各3天给予相应药物3 ml灌胃干预,采用ELISA法测定血清MMP-2、MMP-9及TIMP-1含量。结果与假手术组比较,其余各组大鼠血清MMP-2、MMP-9、TIMP-1升高,差异有统计学意义(P<0.01);与模型组比较,大黄治疗组及大黄素治疗组大鼠血清MMP-2、MMP-9升高,TIMP-1降低,差异有统计学意义(P<0.01)。结论大黄及大黄素可通过促进胰腺细胞外基质的降解,防止胰腺纤维化的形成。

Abstract:Objective To explore the influence of rhubarb and emodin on MMP-2,MMP-9 and TIMP-1 from pancreatic extracellular matrix.Methods SD rats were randomized into four groups: sham-operated group,model group,rhubarb group and emodin group,ten rats in each group.Extracellular matrix sedimengary model was established by pancreaticobiliary duct ligation and caerulein(50 ng/g) injection,and then the rats in different groups were treated with different medicines 3 ml respectively,before and after modeling for 3 days.The MMP-2,MMP-9 and TIMP-1 were detected by ELISA.Results Compared with sham-operated group,the MMP-2,MMP-9 and TIMP-1 were increased in the other groups,with significant difference between them(P<0.01);compared with model group,the MMP-2 and MMP-9 were increased,and the TIMP-1 was decreased in rhubarb group and emodin group,with significant difference between model group and medicine groups(P<0.01).Conclusion Rhubarb and emodin can prevent pancreatic fibrosis by promoting the degradation of the pancreas extracellular matrix.
